CCL20/TNF/VEGFA Cytokine Secretory Phenotype of Tumor-Associated Macrophages Is a Negative Prognostic Factor in Cutaneous Melanoma

چکیده
TAMs constitute a large fraction of infiltrating immune cells in melanoma tissues, but their significance for clinical outcomes remains unclear. We explored diverse TAM parameters clinically relevant primary cutaneous samples, including density, location, size, and polarization marker expression; addition, because cytokine production is hallmark macrophages function, we measured CCL20, TNF, VEGFA intracellular cytokines by single-cell multiparametric confocal microscopy. The Kaplan–Meier method was used to analyze correlation with melanoma-specific disease-free survival overall survival. No significant correlations were observed morphology, or location. Significantly, higher contents the quantified metastasizing compared non-metastasizing skin melanomas (p < 0.001). To mechanistically explore up-regulation, performed vitro studies melanoma-conditioned macrophages, using RNA-seq involved pathways specific inhibitors. show that p53 NF-?B coregulate macrophages. These results delineate pro-oncogenic profile prognostic point combined therapeutic targeting NF-kB/p53 control deviation melanoma.

Background: The aim of sentinel node biopsy (SNB) is to identify subclinical lymph node metastases using a procedure with minimal morbidity, establish more accurate staging in patients with melanoma, determine prognosis, and choose the most suitable treatment in each patient. Objectives: To analyze the outcomes of SNB in patients with cutaneous melanoma, and the impact of this procedure on surv...

متن کاملP157: Periostin Recruits Tumor Associated Macrophages in Glioblastoma Multiform
Glioblastoma multiform (GBM) is the most common and lethal type of primary brain tumors with high rates of morbidity and mortality. Treatment options are limited and ineffective in most of the cases. Epidemiological studies have shown a link between inflammation and glioma genesis. In addition, at the molecular level, pro-inflammatory cytokines released from activated microglia can increa...
متن کاملCutaneous melanoma: prognostic factors.
BACKGROUND Recent data have changed our views of prognostic factors in cutaneous melanoma. While some newer methods have yielded better prognostic information, some insights have evolved as a result of large-scale population-based analyses. METHODS We review current data on several different prognostic factors and divide these factors according to their application in localized primary melano...
متن کاملT-cell activation marker expression on tumor-infiltrating lymphocytes as prognostic factor in cutaneous malignant melanoma.
The central role of T cells in antitumor immunity is well established. However, tumor progression, often seen in the presence of substantial lymphocytic infiltration, suggests that these T cells are not capable of mounting an effective immune response to control tumor growth. Evidence has accumulated that T lymphocytes infiltrating human neoplasms are functionally defective, incompletely activa...
